From the initial characterization of ATP-dependent phosphorylation in the liver by Gene Kennedy in 1954, our understanding of the family has grown from the diverse perspectives of enzymology, biochemistry, biophysics, structural biology, genetics, cellular biology, pharmacology and physiology. Importantly, these advancements have yielded transformative therapeutic approaches to disease, most notably in the field of cancer where kinase inhibition is helping to fulfill the promise of precision medicine.
