Gain a deeper understanding of the fundamental principles underlying recovery/purification operations for biopharmaceutical production—including centrifugation, homogenization, chromatography and ultrafiltration—and how these operations are developed and scaled for use in production. Lectures provide theoretical insight into downstream operations and regulatory expectations for process development. Hands-on lab activities provide experience in experimental methods for determining process parameter ranges and material attributes (if applicable) for the different operations. The course covers material that is especially relevant to scientists and engineers working in downstream biopharmaceutical process development, particularly those new to the field. Its overview of fundamental principles and development methodologies for operations associated with downstream processing also makes the course worthwhile for downstream manufacturing management, process validation staff, and quality assurance professionals.

This intensive laboratory and lecture course will provide participants with the theoretical and practical knowledge to utilize existing and emerging genetic code expansion technology. The laboratory component will focus on troubleshooting GCE tools with attendees and their genes in E. coli expression systems. The workshop will have a demonstration section on expression unnatural protein in mammalian systems with standard controls.
The laboratory component will focus on two common needs in the genetic code expansion field: 1) incorporation of an existing non-canonical amino acid into protein of interest 2) screening a new non-canonical amino acid against a panel of synthetases to determine if the desired non-canonical amino acid can be incorporated with current genetic code expansion tools. As part of the laboratory component of the course, participants will be given the opportunity to bring their research gene of interest and/or their non-canonical amino acid to the workshop to test their expression and incorporation along with optimized controls.
The lecture component will focus on how new genetic code expansion components are generated and attributes of orthogonality, efficiency, fidelity and permissivity are defined, measured and are conditionally dependent. The major applications now afforded by the new non-canonical amino acids will be discussed with a focus on the future of each field and its limitations.

This SRC focuses on bringing together biochemists, geneticists, molecular biologists and clinicians interested in the pathophysiology of the leukemias and lymphomas, and the development of novel therapies for these diseases. Leukemia and lymphoma has provided a number of key paradigms for molecularly targeted cancer therapy, as illustrated by the remarkable success of all-trans-retinoic acid for the treatment of acute promyelocytic leukemia and tyrosine kinase inhibitor therapy for chronic myelogenous leukemia. New and promising small-molecule compounds and antibodies targeting epigenetic regulators, surface receptors and downstream signaling pathways are currently under development and will be discussed at this meeting. Added to these examples of targeted therapy, immunotherapy, including checkpoint inhibition and modified T cell-based therapies have emerged as important future anti-leukemia/lymphoma treatments.
The meeting will cover a broad range of topics, including hematopoietic stem cell differentiation, identification and study of novel oncogenes and tumor suppressors, mechanisms of action of products of chromosomal translocations, normal and pathologic signaling in hematopoietic cells, aberrant epigenetic/transcriptional control, genomics of hematological malignancies, the role of tumor microenvironment, and the development of new experimental models of hematologic malignancies.
