Generation of Monocyte-derived Dendritic Cells (Mo-DCs)

This 2-day training course focuses on hands-on generation of monocyte-derived dendritic cells (Mo-DCs) in theory and practice. Pros and cons of different monocyte isolation strategies are introduced, and new technologies are presented to make monocyte-derived dendritic cells generation more efficient, reliable, and easier than ever. The program includes magnetic cell sorting, cell culture, and immunophenotyping by flow cytometry. Participants will experience how to save resources while still following a well-accepted protocol. The course covers the workflow of Mo-DC generation both in research and in clinical settings.
Course lectures and exercises include:
Introduction to in vitro-generated DCs
Sample handling and isolation of monocytes from blood
Efficient generation of immature and mature Mo-DCs
Microscopic control of Mo-DC generation
Basics of flow cytometry for Mo-DC analysis
Large- and clinical-scale DC generation
Practical laboratory parts include:
Isolation of monocytes from whole blood and from PBMCs
Manual and automated cell isolation methods
Flow cytometric analysis and quality control
Microscopic control of different Mo-DC maturation stages
Flow analysis with the MACSQuant Analyzer
