Small non-coding RNAs (ncRNAs) represent a significant, ubiquitous and ever-growing class of regulatory RNAs (the latest release of the miRBase repository (19.0, August 2012) contains 21,264 miRNA sequences from 193 species; there is no database for other ncRNAs) that are principally involved in control of many cellular processes of most organisms, regardless if bacterial or eukaryotic.

This course will focus on methods enabling characterization of cellular content of ncRNAs as well as providing insights into their function. In particular, we will teach methods allowing investigation of these molecules in all organisms, not only in human or mouse.

The practical part will include ncRNA detection, data analysis, miRNA target prediction, correlation of mRNA and miRNA expression profiles as well as correlation of data obtained by various methods.

During the course we will pay particular attention to following aspects:

a) explanation of different approaches used for ncRNA detection & profiling;

b) detailed troubleshooting for all steps required to characterize ncRNA profiles;

c) comparison of analysis strategies and evaluation of obtained data;

d) importance of standardized approach to miRNA expression profiling and suitable normalization strategies;

e) experimental approaches for the identification of miRNA target and for the functional validation of miRNA activity.
